Identity

Vilon is the dipeptide Lys-Glu (KE), the shortest and earliest of the defined Khavinson peptides. It was derived from Thymalin, the calf-thymus polypeptide fraction, when the originating group set out to identify which components carried the activity of the crude extract.

Molecular target

No receptor. Being only two residues, Vilon is the member of the series for which the direct-DNA-binding hypothesis has been examined most closely, including docking and spectroscopic studies of interaction with specific oligonucleotide sequences.

Signalling pathway

Reported outcomes are transcriptional and epigenetic: changes in expression of genes associated with immune-cell function, and effects on chromatin decondensation in lymphocyte nuclei described in the ageing literature. Its size is both the strongest argument for the nuclear hypothesis, since a dipeptide can plausibly reach the nucleus, and the strongest argument against sequence-specific selectivity, since two residues carry very little information.

Research context

Lymphocyte culture, and rodent lifespan and immunosenescence studies from the originating group, including some of the longest-running experiments in the series. Independent replication outside that group remains limited.
