TB-500 is a synthetic peptide corresponding to the actin-binding region of thymosin beta-4, a 43-residue G-actin-sequestering protein found in most mammalian cells. The supplied material is the acetylated fragment built around the LKKTETQ motif at residues 17-23 of the parent sequence, which is the portion that carries actin-binding activity.
Molecular target
No cell-surface receptor has been established for this fragment. The LKKTETQ motif binds monomeric G-actin directly, forming a one-to-one complex, so the target is a cytoskeletal protein rather than a membrane receptor.
Signalling pathway
Sequestering G-actin shifts the equilibrium between monomeric and filamentous actin. Published work has examined the downstream consequences for cytoskeletal reorganisation, directed cell migration and the transcription of matrix and angiogenic genes, including MRTF/SRF-dependent transcription that is itself sensitive to the free G-actin pool.
