Identity

ARA-290, also called cibinetide, is an 11-residue linear peptide corresponding to the aqueous face of helix B of erythropoietin. That face of the parent molecule is the surface that does not contact the classical erythropoietin receptor homodimer, which is the basis of the whole design.

Molecular target

The innate repair receptor, a heterocomplex of the erythropoietin receptor with the beta-common receptor CD131, the subunit shared with the GM-CSF, IL-3 and IL-5 receptors. Because the peptide does not engage the EPOR homodimer, it is used in models where erythropoiesis and haematocrit change would be confounders.

Signalling pathway

Engagement of the heterocomplex activates JAK2 with downstream STAT3 and PI3K/Akt signalling. The published readouts are anti-apoptotic and anti-inflammatory rather than haematopoietic, studied in tissue under metabolic or ischaemic stress.

Research context

Rodent nerve-injury, ischaemia and inflammation models. Small human trials exist in sarcoidosis-associated small fibre neuropathy, which is unusual for a compound in this catalogue.
