Identity

SNAP-8 is acetyl octapeptide-3, also listed as acetyl glutamyl heptapeptide-3. It is an eight-residue elongation of acetyl hexapeptide-8 (Argireline), and its sequence mimics the N-terminal end of SNAP-25.

Molecular target

The SNARE complex. Rather than binding a receptor, the peptide competes with native SNAP-25 for its position in the ternary SNARE assembly formed with syntaxin and VAMP.

Signalling pathway

A SNARE complex that incorporates the mimetic in place of SNAP-25 is less competent to dock and fuse secretory vesicles. The reported consequence in chromaffin-cell preparations is reduced calcium-dependent catecholamine exocytosis, and the same mechanism is the basis for the compound being studied at the neuromuscular junction.

Bovine chromaffin cell culture and in vitro cosmetic-science assays. The published evidence is in vitro; the two extra residues relative to Argireline were introduced for solubility and formulation behaviour rather than on mechanistic grounds.
