Thymosin alpha-1 is an N-terminally acetylated 28-residue peptide corresponding to the first 28 residues of prothymosin alpha. It was originally isolated from thymosin fraction 5 of bovine thymus, and unlike the crude thymic extracts of the same era it is a single defined sequence.
Molecular target
Toll-like receptors, principally TLR2 and TLR9 on dendritic cells and monocytes. No single high-affinity receptor has been identified.
Signalling pathway
Toll-like receptor engagement recruits MyD88 and activates NF-kappaB. Published work has examined the consequences for dendritic cell maturation, IL-12 and interferon-gamma production and Th1 polarisation, and separately for thymocyte differentiation and natural killer cell activity.
