Vasoactive intestinal peptide is a 28-residue neuropeptide of the secretin and glucagon superfamily, distributed across enteric, central and peripheral neurons and released from a subset of immune cells.
Molecular target
VPAC1 and VPAC2, class B Gs-coupled receptors, with lower-affinity binding at PAC1. The two VPAC subtypes differ in tissue distribution, which is the usual basis for separating peripheral from central effects in a study design.
Signalling pathway
Both VPAC receptors signal through adenylyl cyclase to cyclic AMP and protein kinase A. Three separate literatures draw on this: smooth-muscle relaxation in gut and vascular preparations, circadian pacemaking in the suprachiasmatic nucleus where VPAC2 is required for network synchrony, and immunomodulation, where VIP has been studied for shifting macrophage and T-cell populations toward a regulatory phenotype.
