AOD-9604 is a modified fragment of human growth hormone comprising residues 176-191 of the C-terminus with an added N-terminal tyrosine. It contains none of the receptor-binding regions of the intact 191-residue hormone.
Molecular target
No activity at the growth hormone receptor. The fragment does not bind hGHR, and in the published work it did not raise IGF-1 or reproduce the insulin-antagonistic effects of full-length growth hormone. This dissociation is the entire rationale for the molecule.
Signalling pathway
The lipolytic activity reported in rodent adipose tissue has been attributed to effects on beta-3 adrenergic receptor expression and on hormone-sensitive lipase, but the mechanism remains incompletely characterised and no receptor for the fragment has been identified.
Research context
Rodent adipose tissue preparations and ob/ob mouse models. Human obesity trials did not meet their endpoints and development was discontinued, which is worth knowing when reading the earlier and more optimistic literature.
