Identity

Ipamorelin is the synthetic pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH2, developed after the earlier growth hormone secretagogues GHRP-6 and GHRP-2.

Molecular target

The growth hormone secretagogue receptor GHS-R1a, a Gq-coupled receptor expressed on anterior pituitary somatotrophs and in the hypothalamus. This is the receptor for endogenous ghrelin, and is distinct from the GHRH receptor targeted by sermorelin and CJC-1295.

Signalling pathway

Agonism at GHS-R1a activates phospholipase C, raising inositol trisphosphate and intracellular calcium in somatotrophs. In the animal literature ipamorelin is described as selective because that release occurred without the rise in ACTH, cortisol and prolactin seen with the earlier secretagogues. Because it acts at a separate receptor from GHRH, the two classes are frequently studied together to examine additive release.
