The GHRH receptor, a class B Gs-coupled receptor found on anterior pituitary somatotrophs.
Signalling pathway
Receptor occupancy activates adenylyl cyclase, raising cyclic AMP and activating protein kinase A, which phosphorylates CREB. This drives both immediate release of stored growth hormone and Pit-1-dependent transcription of the growth hormone gene. Because the action is upstream at the pituitary, output in these models remains subject to somatostatin negative feedback, which is the principal design contrast with administering growth hormone directly.
