Identity

PT-141, also called bremelanotide, is a cyclic heptapeptide melanocortin analogue. It is the major metabolite of melanotan II, differing from it by the loss of the C-terminal amide.

Molecular target

The melanocortin receptor family, with the studied activity at MC3R and MC4R and weaker activity at MC1R. The shift away from MC1R relative to its parent compound is the point of pharmacological interest, MC1R being the receptor associated with pigmentation.

Signalling pathway

The melanocortin receptors are Gs-coupled, so agonism raises cyclic AMP and activates protein kinase A. In the rodent literature the effects of interest were localised to hypothalamic and spinal circuits rather than to peripheral tissue, which is why the compound is used in central-nervous-system rather than dermatological models.
