Delta sleep-inducing peptide is the nonapeptide Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, isolated in 1977 from the cerebral venous blood of rabbits during electrically induced slow-wave sleep and named for the EEG band it was associated with.
Molecular target
None identified. Nearly fifty years after isolation no DSIP receptor has been cloned and no consensus binding site established, which is the single most important constraint on interpreting the literature around it.
Signalling pathway
Reported associations with delta-band EEG power, with hypothalamic-pituitary-adrenal axis output and with opioid and GABAergic transmission are correlative. Without a defined receptor there is no established transduction pathway to describe, and competing groups have proposed carrier-mediated and non-specific mechanisms.
Research context
Rabbit, rat and cat EEG studies, largely published between the late 1970s and the 1990s. Results have been inconsistent between laboratories, and the compound is used today mainly as a reference material in sleep-neurochemistry work.
