Cardiogen is the tetrapeptide Ala-Glu-Asp-Arg (AEDR), one of the defined short peptides isolated from the Khavinson series at the St Petersburg Institute of Bioregulation and Gerontology. It is the member of the series assigned to cardiac tissue.
Molecular target
No receptor. The working hypothesis in this literature is direct interaction with DNA: molecular docking and spectroscopic studies describe short peptides of this class binding in the minor groove at specific promoter sequences, and associating with histone proteins, rather than acting at any cell-surface site.
Signalling pathway
The proposed consequence is altered transcription of tissue-specific genes. For Cardiogen the reported readouts are in cardiomyocyte and myocardial preparations, including changes in the expression of contractile and stress-response gene products. The mechanism by which a four-residue peptide would reach the nucleus intact remains the weakest link in this account.
Research context
Cardiomyocyte culture and rodent cardiac models, reported almost entirely by the originating group. The tissue-specificity claim is the part of the hypothesis with the least independent support: several members of the series differ by a single residue yet were assigned to different organs.
