Molecular target

Nicotinamide N-methyltransferase, a cytosolic enzyme that transfers a methyl group from S-adenosylmethionine to nicotinamide. NNMT is highly expressed in adipose tissue and liver.

Signalling pathway

Inhibition has two linked consequences the literature examines separately: nicotinamide that would otherwise be methylated and excreted remains available to the NAD+ salvage pathway, and the SAM methyl pool is spared for histone and DNA methylation reactions.

Research context

Like SLU-PP-332 this is a cell-permeable small molecule, so an oral format is consistent with how the compound is used in the published work — 3T3-L1 adipocyte culture and diet-induced-obese mouse models. Preclinical only.
