Identity

5-Amino-1MQ is 5-amino-1-methylquinolinium, a cell-permeable small-molecule quinolinium salt. It is not a peptide and is supplied as a dry powder.

Molecular target

Nicotinamide N-methyltransferase, a cytosolic enzyme that transfers a methyl group from S-adenosylmethionine to nicotinamide, producing 1-methylnicotinamide. NNMT is highly expressed in adipose tissue and liver.

Signalling pathway

Inhibiting the enzyme has two linked consequences that the literature examines separately. Nicotinamide that would otherwise be methylated and excreted remains available to the NAD+ salvage pathway, and the SAM methyl pool is spared for histone and DNA methylation reactions. Published mouse work using adipocyte NNMT knockdown reported changes in fat mass without a change in food intake.
