Identity

Kisspeptin-10 is the C-terminal decapeptide of the KISS1 gene product. The gene encodes a 145-residue precursor processed to kisspeptin-54; the shorter C-terminal fragments, including this decapeptide, retain full receptor activity, so the ten residues carry the whole pharmacophore.

Molecular target

KISS1R, also catalogued as GPR54, a Gq-coupled receptor densely expressed on gonadotropin-releasing-hormone neurons in the hypothalamic arcuate and preoptic regions.

Signalling pathway

Receptor activation drives phospholipase C, generating inositol trisphosphate and diacylglycerol, mobilising intracellular calcium and activating protein kinase C. The result in slice electrophysiology is depolarisation of GnRH neurons. Because kisspeptin sits one step above GnRH, it is used in models where the pulse generator itself, rather than downstream gonadotropin response, is the variable of interest.

Hypothalamic slice electrophysiology, and rodent and ovine hypothalamic-pituitary-gonadal axis models. Loss-of-function mutations in KISS1R in humans first identified the pathway.
