Selank is a synthetic heptapeptide derived from tuftsin, a naturally occurring immunomodulatory tetrapeptide (Thr-Lys-Pro-Arg) produced by cleavage of immunoglobulin G. As with Semax, a Pro-Gly-Pro extension is added to slow enzymatic degradation.
Mechanism
Research describes anxiolytic activity mediated through the GABAergic system, with reported effects on GABA-A receptor subunit expression. Selank has also been described as inhibiting enkephalin-degrading enzymes, thereby prolonging the action of endogenous enkephalins.
It retains immunomodulatory properties from its tuftsin lineage, including effects on interleukin expression — which is part of why it is studied at the interface of immune signalling and behaviour.
The distinguishing claim
The property that draws research attention is anxiolytic activity without the sedation, motor impairment or dependence associated with benzodiazepines. If that separation holds, the mechanism is worth understanding; the preclinical literature is reasonably consistent on it, and the human literature is again largely Russian.
