KPV is a tripeptide — lysine, proline, valine — corresponding to the C-terminal three residues of alpha-melanocyte-stimulating hormone (α-MSH 11–13).
Why a fragment rather than the whole hormone
α-MSH has two broad activity sets: pigmentary effects via melanocortin receptors, and anti-inflammatory effects. Research on the tripeptide fragment is an attempt to separate the second from the first.
KPV retains anti-inflammatory activity described for the parent hormone while lacking the melanocortin receptor agonism responsible for pigmentation. Notably, its activity appears to be at least partly independent of melanocortin receptors — which is mechanistically interesting, since it suggests a route distinct from classic MC receptor signalling.
Mechanism
The most commonly described action is inhibition of NF-kappaB signalling, the master transcriptional regulator of inflammatory gene expression, with downstream reduction in proinflammatory cytokine production. Work in intestinal epithelial models has examined uptake via the PepT1 transporter.
Research status
Preclinical and in vitro, with a focus on mucosal and intestinal inflammation models. Human clinical data is minimal.
