This preparation combines two compounds that raise growth hormone by different, complementary routes. Understanding why they are paired requires seeing that they act on separate receptors.
CJC-1295 without DAC
CJC-1295 no-DAC — also called modified GRF(1–29) — is an analogue of the first 29 residues of growth hormone-releasing hormone, the biologically active portion. Four amino acid substitutions resist enzymatic degradation, particularly by DPP-4.
The “no DAC” distinction matters. The DAC (Drug Affinity Complex) version binds serum albumin, extending half-life to days and producing a sustained elevation. Without DAC, the half-life is roughly 30 minutes, so it produces a pulse rather than a plateau — closer to physiological secretion.
It acts at the GHRH receptor on pituitary somatotrophs.
Ipamorelin
Ipamorelin is a pentapeptide agonist at the growth hormone secretagogue receptor (GHS-R1a) — the ghrelin receptor. This is an entirely separate receptor from the GHRH receptor.
Its defining property is selectivity. Earlier secretagogues also raised cortisol, prolactin and ACTH. Ipamorelin was developed to stimulate growth hormone release with minimal effect on those, which is why it largely replaced older compounds in research use.
Why combine them
The two receptors converge on the same cell but through different signalling. GHRH receptor activation raises cyclic AMP; GHS-R1a signals through the phospholipase C pathway. Stimulating both simultaneously produces a greater release than either alone — a synergistic rather than additive effect.
Both preserve pituitary feedback, so IGF-1 mediated negative feedback remains intact, unlike direct growth hormone administration.
